LRIG1 / LIG-1蛋白背景綜述
Mouse Leucine-rich repeats and immunoglobulin-like domains protein 1, also known as LIG-1 and LRIG1, is a single-pass type I membrane protein which is widely expressed. LRIG1 / LIG-1 contains 3 Ig-like C2-type (immunoglobulin-like) domains, 15 LRR (leucine-rich) repeats, 1 LRRCT domain and 1 LRRNT domain. LRIG1 / LIG-1 act as a feedback negative regulator of signaling by receptor tyrosine kinases, through a mechanism that involves enhancement of receptor ubiquitination and accelerated intracellular degradation. LRIG1 / LIG-1 is an endogenous inhibitor of growth factor signaling and a proposed tumor suppressor. LRIG1 / LIG-1 may be overexpressed in prostate cancer. LRIG1 / LIG-1 is an important determinant of prostate cancer growth, and the implications of its expression on patient outcome depend on the clinical and biological circumstances. LRIG1 / LIG-1 evolved in gliomas as a rare feedback negative attenuator of EGFR and could offer a novel therapeutic target to treat patients with malignant gliomas.
